Background & Objective: There is a fast growing tendency in the consumption of herbal remedies in developing countries. One of the traditional medicines used for male infertility is Date Palm Pollen (Phoenix Dactylifera) (DPP). The goal of the present study is to investigate the effect of aqueous extract of DPP on the rates of in vitro viability and proliferation in neonate mouse Spermatogonial Stem Cells (SSCs). Materials & Methods: Cell suspension including sertoli cells and SSCs were isolated from testes of six to 10 day-old mice by two steps enzymatic digestion. The cell suspension was cultured in Dulbecco's Modified Eagle's Medium (DMEM) with or without 0.06, 0.25, and 0.62 mg/ml of aqueous extract of DPP during two weeks. In order to evaluate the rate of SSCs growth at the end of culture, the total number of the cells and living cells were considered as proliferation and survival rates, respectively. Results: The results showed that there were no significant difference between the percent of viability and proliferation in control and 0.06, 0.25 and 0.62 mg/ml of DPP-treated groups (P > 0.05). Conclusion: Our study showed that treatment of neonatal mouse testicular cell suspension with DPP had no toxic effects on viability percent and the proliferation rate of these cells. Therefore, we can use DPP to evaluate the in vitro pattern of SSCs colonization in the future studies. [ABSTRACT FROM AUTHOR] |
