Abstrakt: This protocol chapter outlines a guide for investigating ghrelin receptor activation of Gq signaling using Bioluminescence Resonance Energy Transfer (BRET). Focusing on the eBRET 2 and NanoBRET™ variants, the chapter covers steps from cell culture to transfection, ligand stimulation, and BRET measurements, offering a robust protocol to examine the temporal aspects of Gq signaling in live cells. This methodology facilitates a nuanced understanding of Gq-mediated cellular responses, but also provides researchers with a valuable tool for dissecting other related signaling pathways.
