In the order to determine the growth and changes of NADH and other substances during the process of fermentation and ensure the quality and stability of the substance, it is prudent to have real-time monitoring. The main purpose of the current research is to measure the fluorescence response of the NADH substance in the fermenting broth. Measuring NADH requires an excitation light source in the 340 nm band and a receiving end requiring a 450 nm band receiver. Measurement methods include: 1). A light excitation part using a specific wavelength matched with a MCU that sends out the PWM cycle of the signal to drive the brightness of the LED. 2). The light receiver part uses a diode with a specific wavelength matched with a special optical isolation filter film that limits the light emitting and receiving wavelength within the specified limits. In addition, a MCU is used to perform digital signal processing and results are presented on a LCD while keeping track and recording changes. The design of the probe for the emitting and receiving end is an L-shaped steel tube in order to reduce the distance between the two ends. This reduction in distance is to prevent a power loss due to distance in the optic fiber. Initial measurements made use of fluorescent agents to simulate the fermentation broth of the NADH substance. It also serves as the preliminary measurement and ground in choosing appropriate amplification options. Follow-up test and comparison on NADH fluorescence measurement made use of RO water and Sigma N8129 NADH that is mixed to form 9 different levels of concentration. Furthermore, 2 kinds of broths are separately investigated under fluorescence spectrometer and photoelectric sensor circuit passing through digital signal conversion are read, analyzed and compared.
