Determination of Urinary Cotinine of Elementary Students and Communities by HPLC-UV Method

An HPLC with UV detection have established for analysis of cotinine in urine. Mobile phase was consisted of acetate buffer and methanol(70 : 30, v/v). The cotinine was separated by Merck RP-18 column (250-4 mm, 5 μm). The wavelength of UV detector was set at 260 nm. The regression equations were linear between 5 ~ 8000 ng/mL (R2 > 0.999). This method was applied for determination cotinine contents in urine of elementary school students, community residents and smokers. (1) The sample source of students are come from Kaohsiung elementary school. The exposure to SHS of students of fourth, fifth and sixth grades in an elementary school was studied. 882 effective questionnaires were collected. The urine samples actually are 126. In part of number of smokers at home, four groups were separated into as following: zero, one, two and three, the mean contents of cotinine (ng/mL) of above four groups are (7.40), (7.17), (7.63), (8.55), respectively. In part of place of exposure, four groups were separated into as following: at home, at workplace of parents, at remedial class and exposure to ETS environment, the mean contents of cotinine (ng/mL) of above four groups are (7.26), (8.96), (6.60), (7.52), respectively.The mean contents of cotinine (ng/mL) of non-exposure to SHS at home and workplace of parents and remedial class is (6.40). (2) The sample source of community residents was from eight community. The mean contents of cotinine of smokers is 1.61.16 ng/mL. The mean contents of community residents that non-exposure to second hand smoke (SHS) at home or/and workplace is 16.55 ng/mL. The mean contents of community residents that exposure to second hand smoke (SHS) at home is 16.10 ng/mL. The mean contents of community residents that exposure to second hand smoke (SHS) at workplace is 24.34 ng/mL (3) Have 44 smoker’s sample. The mean age is 27.59. The mean score of Fagerstrom Tolerance Questionnaire is 5.32. The smoker’s contents of urinary cotinine correlate with the number of cigarettes per day, the smoking behaviour, the grade of Fagerstrom Tolerance Questionnaire. (Pearson''s correlation coefficient >0.570 )(4) A voluntary went reduce his number of smoke. 51 years old.The item of satisfactional questionnaire is reduce at Energy/fatigue and General health. The ideal number of smoke and the actually number of smoke smaller than 1.93 every day. The regression equation have good credibility.
