The effects of Lonicera japonica extracts on melanogenesis

In this study, two active compound caffeic acid and chlorogenic acid on tyrosinase activity were isolated from Jinyinhua extract by the process of activity-assay fractionation and column chromatography. These structure were elucidated by spectroscopic methods. Their potential effect on melanogenesis were examined in murine melanoma B16F1 and A2058 cells. The tyrosinase activity were apparently elevated in cells by treating with the Jinyinhua extract or each active compounds. While caffeic acid exhibited more potent than chlorogenic acid. Moreover, the Jinyinhua extract or isolated components could directly stimulate the activity of tyrosinase in test tube. In addition, treatment with Jinyinhua extract and the active component markedly increased the melanin content of B16F1 cells in a dose-dependent manner. We analyzed the protein level of tyrosinase by Western blot using anti-tyrosinase antibody. According to the raise of tyrosinase potein responsive to the treatment of Jinyinhua extract. Treatment of 1.5 % (w/w) Jinyinhua extract enhanced the melanin index on the surface of human skin, leading to the skin darkening. Summary, the active ingredients isolated from Jinyinhua can act as tyrosinase stimulants. Our findings provide evidence to show that Jinyinhua extract and caffeic acid have the potency to be a therapeutic or cosmetic ingredient in treatment with melanin deficiency symptom.
