FFPE tissue is the standard by which biopsied tissue is preserved for molecular analyses, such as NGS. However, current methods to purify nucleic acid from FFPE are technically challenging, often limiting both the yield and quality obtained. We describe here the results using an automated bench-top system to perform isotachophoresis; a novel method for nucleic acid purification. The Agilent QC Plex assay was used to assess the amplifiable yield of each FFPE sample for amplicon based NGS. NGS libraries were enriched using an Agilent SureMASTR assay and sequenced on the Illumina MiSeq sequencer. |
