Popis: Noscapineisanaturalalkaloidthatisusedasanantitussivemedicine.However,italsoactsasaweakanticanceragentincertaininvivomodelsthroughamechanismthatislargelyunknown.Here,weperformedstructuralstudiesandshowthatthecytotoxicagent7A-O-demethoxy-amino-noscapine(7A-aminonoscapine)bindstothecolchicinesiteoftubulin.Wesuggestthatthe7A-methoxygroupofnoscapinepreventsbindingtotubulinduetoastericclashofthecompoundwiththeT5-loopofa-tubulin.Wefurtherproposethattheanticanceractivityofnoscapinearisesfromabioactivemetabolitethatbindstothecolchicinesiteoftubu-lintoinducemitoticarrestthroughamicrotubulecytoskeleton-basedmechanism.
