A tumor-associated antigen isolated from human breast adenocarcinoma

Popis: Breast tumor antigen (BTA) was isolated from human adenocarcinoma obtained from a single patient. The BTA was solubilized either with hypertonic salt or with buffered detergents and purified by three-step ammonium sulfate fractionation, anion exchange chromatography, and preparative polyacrylamide disc gel electrophoresis (PAGE). About 15 mg BTA was isolated from 75 g wet tumor tissue from a single donor. Similar studies with normal human breast, lung, liver, and splenic tissues failed to reveal the presence of BTA or closely related molecules. The purified BTA is a glycoprotein (33% CHO), migrates as a single band on sodium dodecyl sulfate-PAGE, and has an estimated MW of 67,000. Its mobility was different from those found for carcinoembryonic antigen, α-fetoprotein, or donor histocompatibility antigens. The BTA is stable at −20°C and may be readily isolated in the homogeneous form for additional studies.
