Popis: | Summary In vitro synthesis of glutamic oxaloacetic transaminase isozymes of rat liver was studied using a rabbit reticulocyte protein-synthesizing system, polyribosomes of rat liver and specific antibodies against cytoplasmic and mitochondrial isozymes. Sodium dodecyl sulfate polyacrylamide gel electrophoresis showed that the cytoplasmic and mitochondrial isozymes synthesized in vitro had molecular weights which were respectively similar to, and larger by about 2,000 daltons than, those of the corresponding subunits of the mature enzymes. These mature enzymes competed effectively with the in vitro products for interaction with the corresponding antibodies. A mitochondrial membrane preparation from rat liver converted the mitochondrial isozyme synthesized in vitro to the mature form. |
