Popis: | Pregnant Dutch rabbits were treated with either 1 or 3 mg/kg of 6-AN intraperitoneally as a single injection during the preimplantation period (Day 1, 3, or 6) or during the crucial period of organogenesis (Day 9 or 12). Those treated prior to implantation had a low conception rate and a high mortality rate (resorptions) of the early embryo, but lack of gross or skeletal teratogenic effect on the viable kits. Litter and fetal size were unaffected. When 6-AN was injected during organogenesis, there was a marked dose- and time-related teratogenic effect. The litters at term were characterized by an increased incidence of nonviable fetuses, and in the viable kits, a reduced size. The kits had severe anomalies including a high frequency of gross and/or skeletal defects consisting of aphakia or microphthalmia, cleft lip and palate, tail defects, and retarded ossification or malformations of the skull, vertebral column, sternabrae, ribs, limbs, and phalanges. The teratogenic syndrome most closely paralleled that of the albino rat among the other susceptible species. |
