Detection of Nontuberculosis Mycobacteria in National Reference Laboratory (NRL) of Uzbekistan

Popis: Introduction: In Uzbekistan, the incidence of tuberculosis (TB) decreasing, from 52 per 100 thousand people in 2011 to 42 in 2018. In recent years, cases of detection of NTM have begun to be recorded. Aim and Objectives: To analyze the frequency of occurrence and species diversity of NTM Methods: 14431 diagnostic samples were analyzed in 2019. Diagnostic materials (sputum, BALF, pleural fluid, etc.) obtained from patients were examined in NRL at The Republican Specialized Scientific and Practical Medical Center of Phthisiology and Pulmonology. The following research methods were carried out: smear microscopy, culture [Middlebrook 7H9 liquid nutrient medium in a BACTEC MGIT 960 automated system (BACTEC™MGIT™960 System, Becton Dickinson, USA)], MPT64 identification test (SD Bioline TB Ag MPT64 test, Korea). The NTM species was determined by GenoType Mycobacterium AS/CM DNA-strips version 1.0 (Hain Lifescience GmbH, Germany). Results: A study of 14431 samples of diagnostic material was taken in 2856 (19.7%) (with a positive growth and lack of contamination). Among 2856 samples of a positive cultural result of the MPT64: 2845 (99.6%) cultures of the Mycobacterium tuberculosis complex and 11 (0.4%) NTM cultures were obtained. 11 NTMB cultures are aimed at species identification by DNA strip hybridization. GenoType Mycobacterium AS/CM, where 5 types of NTM have been identified (MAC-5 (M.avium ssp. - 3, M.intracellulare - 2); M.gordonae - 4; M.chelonae - 1, M. kansassi - 1). Conclusions: Using the full potential of the laboratory will increase the identification of NTM. The data indicate that research on this issue should be continued.
