Popis: This protocol describes the acquisition of Raman spectra on formalin-fixed paraffin-embedded biological tissue with the following characteristics: 4 µm tissue section placed on a glass slide (H&E-IHC stained-slide for tissue visualization) 4 µm tissue section placed on an aluminum slide (Raman micro-spectroscopy) rapid dewaxing Raman micro-spectroscopy: 5 accumulations of 10 seconds, 50x short working distance objective (N.A. 0.75) Of note: this protocol addresses the Raman micro-spectroscopy slide preparation but does not fully describe the H&E-IHC slide used for tissue visualization (see Grosset et al, Appl Immunohistochem Mol Morphol, 2019)
