Poster presented at the 2022 NIH-funded 4D Nucleome Network Annual Meeting. 12/6-8/2022 San Diego, CA, USA (https://www.4dnucleome.org/4dn-annual-meeting-2022/) Abstract: A key output of the 4DN project is the open publication of datasets related to the structure of the human cell nucleus and the genome Recent years have seen a rapid expansion of FISH omics methods, which quantify the spatial organization of DNA, RNA, and protein in the cell and provide an expanded understanding of how higher order chromosome structure relates to transcriptional activity and cell development Despite this progress, FISH based image data are not yet routinely published because of the lack of standard specifications for data exchange The 4 DN Imaging Working Group ( and the 4DN Data Coordination and Integration Center ( collaborate with the Integrating Imaging and Omics Working Group ( to develop community specifications for image data formats, quality control, and documentation metadata Microscopy Metadata specifications and tools developed by the IWG have recently been published 1 2 and are presented in a related poster Here we present the 4 DN FISH Omics Format Chromatin Tracing FOF CT a community data standard for processed results deriving from Chromatin Tracing techniques, including ORCA, MINA, Hi M, OligoFISSEQ, DNA MERFISH, DNA seqFISH+, IGS To demonstrate the value of FOF CT, we also present a collection of Chromatin Tracing Datasets that were recently deposited to the 4 DN Data Portal and are ideally suited for promoting reuse, further processing, and integrative modeling An extension format for genomic Single Molecule Localization Microscopy ( FISH omics techniques, such as OligoSTORM, is under development and will be presented at a later date [https :://doi org/ 10 1038 /s 41592 021 01315 z 2 https :://doi org/ 10 1038 /s 41592 021 01327 91] |
