Adenosinetriphosphatase systems have been isolated from renal cortex of several species. In guinea pig preparation, the “membrane ATPase,” or Na + -K + -Mg ++ -dependent and ouabain-sensitive component comprises 80–85% of total activity, and is highly specific as regards both substrate and inhibitors. It is inhibited only by heavy metals, cardiac glycosides and certain of the acylphenoxyacetate series of diuretics. Consistent qualitative and quantitative correlations between inhibitory activity in vitro and pharmacological activity are evident, and structure-activity relationships are discussed. |
