IMPLEMENTATION OF SSR MARKERS IN MANGO BREEDING IN AUSTRALIA

Popis: The routine use of molecular markers to assist in varietal identification and selection was identified as a means of improving breeding efficiency in The Australian Mango Breeding Program. Knowledge of the extent of genetic diversity is a major consideration in designing mango breeding strategies. Eleven microsatellites or simple sequence repeat (SSR) markers were evaluated for their usefulness to identify varieties, validate progeny and parents, and estimating genetic diversity of populations. The SSR markers were tested on a diverse range of Mangifera indica L. cultivars, several related Mangifera species from the Australian National Mango Genebank (ANMG), and select parents and progeny of the National Mango Breeding Program. The markers proved ideal for fingerprinting varieties, with an average of 8.36 alleles per locus identified to distinguish between all 105 accessions tested. The markers were used to determine the genetic diversity of accessions in the ANMG, identifying duplications or incorrectly named accessions. Among breeding progeny, SSR markers identified the parents of hand- and open-pollinated mango hybrids with 95% confidence. The successful application of molecular markers for genetic fingerprinting, validating parents and progeny and estimating genetic diversity will greatly improve breeding efficiency in mango.
