A simple modification of the micro-electrophoresis procedure for separation and identification of hemoglobin variants is described. Changing the buffer from barbital pH 8.6 to Tris-glycine pH 9.3 at the same ionic strength increases the negative charge of the hemoglobins, and provides an environment of positively-charged counter ions (glycinium) for them, thus increasing their relative mobility and resolution. The method is of value in detecting hemoglobinopathies which otherwise may be missed. The procedure is suitable for routine clinical hospital laboratories and is a valuable research tool for the rapid screening of samples in studying the genetics of hemoglobin synthesis. |
