Sartorius (formerly ForteBio) SPR system is highly sensitive to measure the binding kinetics of small molecule fragments. Important factors responsible for successfully screening small molecule fragments lie with the proper sample preparation before injecting into the SPR system. One of the major limitations of the HTS campaign using SPR with small molecules is solubility. Fragments are more soluble than small molecules. However, if even one or a small fraction of fragments in a screening plate are less soluble, it can lead to blockage of SPR system capillaries. This chapter provides optimal details required for the preparation of fragments to minimize clogging. |
