A flow injection analysis (FIA) system was designed for the determination of free and liposome-released peroxidase-enzyme activity. The inclusion of an extra washing channel containing 0.1% Triton X-100 in the FIA system provided a solution to problems of electrode fouling and slow recovery of response. The electrode recovery rate is three times higher than with the wash solution without Triton X-100. Based on 16 replicates of the solution containing 3 mU enzyme, the relative standard deviation of the enzyme determination was found to be 1.5%. Based on the sensitivity obtained with this prototype FIA system, it should be possible to adapt this approach for an immunochemical-based flow injection analysis system capable of measuring an analyte at the femtomole level. |
