In the present paper we propose the design and realization of an experimental device of Langendorff type, controlled in pressure and with the vibrating column of fluid, used for decellularization of the rat heart with a surfactant solution of sodium dodecyl sulfate (SDS). The experimental device consists of two constructive parts: a hydraulic-electronic part that ensures constant pressure for retrograde heart perfusion and an electromagnetic assembly that induces the vibration of the liquid column in the immediate vicinity of the device cannula with a frequency of 15 Hz. The decellularization solution is recirculated during the experiment, so that by serial spectrophotometric determinations of the concentration of deoxyribonucleic acid (DNA) and proteins in this solution can determine the kinetics of decellularization and the moment when it is completed. |
