Activation of NF-kB transcription factor in hepatoma cell cultures by carcinogenic polycyclic aromatic hydrocarbons

Popis: | The activation of NF-kB transcription factor produced by carcinogenic polycyclic aromatic hydrocarbons (PAHs) benzo/a/pyrene (BP) and 3-methylcholanthrene (MC) in cells with (human hepatoma HepG2) or without (rat hepatoma G27) the expression of the Ah receptor has been studied. The NF-kB activity was estimated by NF-kB-dependent I-kB gene expression and by the transcriptional activity of the luciferase reporter gene under the NF-kB-sensitive promoter. BP and MC weakly enhanced the NF-kB activity in proliferating Hep G2 cells, whereas the stimulation of NF-kB activity was significantly higher in resting cells. The activation of NF-kB by BP and MC in hepatoma G27 cells was significantly higher than in Hep G2 cells in both proliferating and resting cells. The role of Ah receptor in the PAH activation of NF-kB transcription factor is discussed. |
