Live imaging of the entire process of mechanical force transmission and cell fate specification in mammalian blastocysts has not yet been achieved. Here, we present a protocol for live imaging and genetic reporting of mechanical forces in cell fate control of mouse blastocyst. We describe steps for mouse embryo microdissection, mounting, and live imaging. We also detail a transformative means to precisely track cell histories over space and time, containing data pre-processing and cell lineage assignment. |
