Spectrofluorometric Determination of Substituted Dihydroisoquinolines and Tertiary Tetrahydroisoquinolines as Their Isoquinolinium Derivatives

Abstrakt: A spectrofluorometric method was developed for the determination of a substituted dihydro- and a tertiary tetrahydroiso-quinoline in biological fluids based on a chemical and/or photochemical oxidation reaction to produce the isoquinolinium derivatives which fluoresce strongly in acidic media. This principle was used for the development of assays of these compounds in blood and urine with sensitivity limits in the submicrogram range. TLC was used to establish the specificity of the assays for the intact drugs and their biotransformation products. The method was applied to the determination of blood levels and the urinary excretion of these compounds administered in the dog and in man. Both compounds were found to be rapidly absorbed and extensively metabolized by both species.
