Brucellosis is a disease caused by bacteria of the genus Brucella. Brucella abortus infects cattle, causing economic losses due to abortions, deaths of calves, decreased milk production, loss of national and international markets and human disease. In Argentina, Corrientes and Formosa provinces there are 8 million hectares suitable for the breeding of buffaloes (Bubalus bubalis); in this area bovine and buffaloes are breed together. The fact that B. abortus is able to infect buffaloes was demonstrated in many countries. The aim of this study was to determine the cause of an outbreak of abortions occurred in a herd of buffaloes in Formosa, Argentina and the prevalence of brucellosis in this herd. 462 buffaloes were bled. Sera were frozen at - 20 °C until processing. The techniques used were: Plate Buffered Antigen (BPA), Complement Fixation (CF), Competitive ELISA (cELISA), Serum Agglutination Test (SAT) and 2-Mercaptoethanol (2-ME). These tests were performed as following international recommendations. Fluorescence Polarization Assay (FPA) was processed following the methodology described by Nielsen et.al. Samples of aborted fetus were collected such us abomasums contents and organs (liver, spleen, kidney, and lung) for bacterial isolation. The culture media used were blood agar, nutrient agar and Skirrow. Cultures were carried out at 37 °C in aerobic and microaerophilic atmosphere for 7 day. Kappa test was applied to measured concordance among the different techniques. The serological results indicated a prevalence of 36%, 35%, 42%, 41%, 27% by BPA, FC, FPA, c-ELISA and SAT and 2-ME respectively. The analysis using Kappa indicated a concordance of 95% between BPA and FPA Kappa: 0.773 (p<0.00001). A strain of B. abortus was isolated and classified as biotype 1. We concluded that the infection in de herd and the abortions were due to Brucella abortus. According to our results BPA test is considered useful as a screening test and FPA as confirmatory test for buffalo. New trials should be made to know the importance of buffaloes in the maintenance of infection in bovine. [ABSTRACT FROM AUTHOR] |
