Glioblastoma remains an incurable form of brain tumor. Existing therapeutic methods can only produce slight prolongation of the lives of patients with this diagnosis. There is therefore a need to seek new approaches and develop new methods of treating glioblastoma. We describe here the principle of an approach to glioblastoma tumor cells which consists of targeted inhibition of the proliferation of L1CAM-positive cells using aptamers. L1CAM is regarded as a marker of gliomastem cells, whose presence in a tumor may be responsible for resistance to treatment. The studies reported here led to selection of the yly12 aptamer from a panel of aptamers for L1CAM; this aptamer was shown to have antiproliferative effects which were more powerful in the case of human glioblastoma cells with elevated L1CAM expression. Thus, this approach may solve the issue of glioblastoma cell resistance and prevent tumor recurrence by acting on glioma stem cells. [ABSTRACT FROM AUTHOR] |
