The synthesis of bicyclo[7.4.1]enediynes 2a and 2b are described. The bicyclo[7.4.1]enediyne 2b was found to be a poor DNA cleaver but did cause damage to several protein isolates.

A series of electronically and sterically altered enediyne cores, based on esperamicin, were prepared using a cobalt mediated cyclization strategy in an attempt to improve the antitumor activity of this class of compounds. The biological activity of

The simple enediyne core molecule 1 is shown to cause protein damage to several cellular protein isolates in vitro. The aromatization of 1 to generate a benzene diradical under simulated physiological conditions is also described.

Maduropeptin (MDP) is a recently isolated antitumor antibiotic, consisting of an enediyne-containing chromophore embedded in a highly acidic protein. This holoantibiotic damages duplex DNA in vitro, producing a mixture of single- and double-strand br
