We have cloned and sequenced the DNA of alkaliphilic Bacillus firmus OF4 encompassing the cta operon that encodes a pH-regulated cytochrome caa3 oxidase. The gene organization is identical with that of the homologous Bacillus subtilis caa3 oxidase lo

The terminal oxidase content of Bacillus firmus OF4, a facultative alkaliphile that grows well over the pH range of 7.5 to 10.5, was studied by difference spectroscopy. Evidence was found for three terminal oxidases under different growth conditions.

Two protonophore-resistant mutants, designated strains CC1 and CC2, of the facultative alkaliphile Bacillus firmus OF4 811M were isolated. The ability of carbonyl cyanide m -chlorophenylhydrazone (CCCP) to collapse the protonmotive force ( Δ μ ¯ H

The bioenergetics of two extreme halophiles, Halobacterium halobium and Halobacterium marismortui, have been investigated by respiratory studies and in vivo nuclear magnetic resonance (NMR) spectroscopy. The phosphorus resonances of both species were

The chemiosmotic model of energy transduction offers a satisfying and widely confirmed understanding of the action of uncouplers on such processes as oxidative phosphorylation; the uncoupler, by facilitating the transmembrane movement of protons or o

Transhydrogenase couples the transfer of hydride-ion equivalents between NAD(H) and NADP(H) to proton translocation across a membrane. The enzyme has three components: dI binds NAD(H), dIII binds NADP(H) and dII spans the membrane. Coupling between t

The Tyr residue in the mobile loop region of the soluble, domain I polypeptide (called Ths) of the proton-translocating transhydrogenase from Rhodospirillum rubrum has been substituted by Asn and by Phe. The recombinant proteins were expressed at hig

Tn 917 transposition libraries prepared from Bacillus subtilis were screened for mutants that had insertions in the chromosome resulting in resistance to the protonophore carbonylcyanide m -chlorophenylhydrazone (CCCP). Five such strains were charact

An 1100-bp DNA fragment cloned from alkaliphilic Bacillus firmus OF4 contained an open reading frame deduced to encode a 54-amino-acid, glutamine-rich protein with 35.6% identity to Bacillus subtilis small, acid-soluble spore protein-gamma (SASP-gamm
