Aggrecan is one of the first proteins to be depleted from articular cartilage in early osteoarthritis. We investigated the molecular differences between matrix metalloproteinase (MMP)- and aggrecanase-mediated aggrecan degradation, as a consequence o

Objective To identify, characterize, and compare proteolysis peptide products generated by metalloprotease digests of human articular cartilage. Methods Human articular cartilage was digested by the addition of exogenous metalloproteases, including m

Summary Objective Aggrecan is the major proteoglycan in articular cartilage and is known to be degraded by various proteases, including matrix metalloproteinases (MMPs). The present study was undertaken to develop immunoassays detecting aggrecan and

The degradation of extracellular matrix (ECM) components of articular cartilage is a hallmark of joint-destructive diseases like osteoarthritis (OA) and rheumatoid arthritis (RA). A major component of the ECM is aggrecan, a proteoglycan, which is amo

Emerging evidence supports the concept that biochemical markers are clinically useful non-invasive diagnostic tools for the monitoring of changes in cartilage turnover in patients with destructive joint diseases such as osteoarthritis (OA) and rheuma

Background Rheumatoid arthritis (RA) is a chronic auto-immune disease with extensive articular cartilage destruction. Aggrecan depletion, mediated by aggrecanases is one of the first signs of early cartilage erosion. We investigated, whether measurem

Purpose: Aggrecan is the predominant proteoglycan in the extracellular matrix of articular cartilage. One of the early steps in initiation of cartilage degradation is loss of proteoglycans, which leads to further degradation of the collagen network.
