Extraembryonic tissues contribute to animal development, which often entails spreading over embryo or yolk. Apart from changes in cell shape, the requirements for this tissue spreading are not well understood. Here, we analyze spreading of the extrae

To better understand the interactions between biological molecules, a high optical resolution in all three dimensions is crucial. The intrinsically lower axial resolution of microscopes however, is a limiting factor in fluorescence imaging, correspon

Evolutionary novelty arises from gradual and continued changes rather than disruptive transformations. Paleontological evidence for gradual divergence has been provided by the discovery and analyses of missing link fossils. Analogous principles of gr

Understanding the cellular response to DNA strand breaks is crucial to decipher the mechanisms maintaining the integrity of our genome. We present a novel method to visualize how the mobility of nuclear proteins changes in response to localized DNA d
This chapter introduces the principles and advantages of selective plane illumination microscopy (SPIM) and compares it to commonly used epifluorescence or confocal setups. Due to the low phototoxicity, speed of imaging, high penetration depth, and s

We employ near-infrared femtosecond pulses to probe local alterations of chromatin dynamics caused by DNA strand breaks. Nonlinear confocal nanomanipulation enables us to visualize how DNA damage signaling emanates spatio-temporally in a live cell.
