DNA damage inhibits elongating RNA polymerase II, but also initiates genome-wide transcriptional responses. Here the authors reveal that particularly promoter-bound Pol II is degraded upon DNA damage in a GSK3 signaling-mediated response.

Summary: Transcription-replication (T-R) conflicts cause replication stress and loss of genome integrity. However, the transcription-related processes that restrain such conflicts are poorly understood. Here, we demonstrate that the RNA polymerase II

The ten-subunit transcription factor IIH (TFIIH) plays a crucial role in transcription and nucleotide excision repair (NER). Inactivating mutations in the smallest 8-kDa TFB5/TTDA subunit cause the neurodevelopmental progeroid repair syndrome trichot

Faithful transcription of eukaryotic genes by RNA polymerase II (Pol II) is essential for proper cell function. Nevertheless, the integrity of the DNA template of Pol II is continuously challenged by different sources of DNA damage, such as UV-light,

Landsverk et al. show that the RNAPII S5 phosphatase complex WDR82/PNUTS-PP1 suppresses replication stress. WDR82/PNUTS-PP1 promotes degradation of RNAPII on chromatin, thereby reducing the residence time of RNAPII. Their results suggest that proper
